I an flying to Nashville in November and will have a 1hr 40min layover in Charlotte, but no plane change. Will we be allowed to exit the plane for a brief period of time and if so, what happens to our seats.

Yes you can get off. You would get back on the plane before they board the new passengers.

On that last part, you'll probably want to leave something on your seat to show that it is taken, just in case you are delayed getting back to the plane for whatever reason. They will usually allow you to get off and back on with a driver's license.
